Single-molecule sequencing of immunopeptides and antigenic protein markers on the Platinum® instrument

Single-molecule protein sequencing offers the sensitivity needed to explore the immunopeptidome and profile tumor-associated antigens. We demonstrate the use of the Platinum benchtop instrument to sequence immunopeptides and proteins at the single-molecule level. Peptides are loaded into nanoscale chambers on a chip, where dye-labeled N-terminal amino acid recognizers bind and generate distinct kinetic patterns. Aminopeptidases sequentially expose amino acids for recognition, and kinetic signatures are analyzed to determine peptide sequences. This process can identify amino acid variants or post-translational modifications, crucial for understanding immune checkpoints and functions, enabling advances in immune cell health and therapy.
